Abstract:

SUMMARY RESEARCHES ON LOGGERHEAD TURTLE Caretta caretta (L. 1758) GENETICS AT TURKISH MEDITERRANEAN NESTING BEACHES In this study, 266 from 11 important nesting beaches in Mediterranean coasts in Turkey and one from Çanakkale, totally 267 dead turtles were sampled. And genetic materials were obtained from these samples to study on Caretta caretta genetics. This research, three extraction methods were used comparatively to purify DNA from muscle tissues. It's estimated that; the DNA extracted from muscle tissues with Chelex® 100 is highly-purfied and the processes are taster, easier, cheaper and the risk of contamination is lower than "Wizard" and "Phenol-Chloroform" extraction methods. Extraction using Chelex® 100 is seen as an alternative method in order to obtain higly- purified genetic material for further observations on both mtDNA and nDNA loci of Caretta caretta.
