Biocompatible carriers containing vitamin D and PRP provide remarkable clinical improvement on bone defects 

Abstract

The aim of this study is to investigate the effects of biocompatible carriers containing vitamin D and PRP on bone defects created in rat calvaria. Fifty-two male Sprague Dawley rats were used and divided to six groups i.e.  Calvarial defects were examined by sacrificing the rats in equal numbers at the 1st, 2nd, 4th and 8th weeks. The newly formed bone was evaluated with histopathologic, microtomographic, macroscopic methods as well as the biochemical analysis was performed in the plasma samples. Immunhistochemical expression of FGF23, VDR, RANK biomarkers, which determine bone formation and vitamin D receptor, were investigated. The amount of ossification, bone volume and mineral densities were found to be significantly higher in the 4th and 8th weeks as the biocompatible material delivers with calcitriol. The histological and macroscopic examinations revealed that the defect area was formed with new cells and the defect area shrank in “calcitriol + chitosan + PRP” group. The expression of RANK, FGF23, VDR antibodies was more and more intense in the “calcitriol + chitosan + PRP” group as compared to other groups and control experiment. In conclusion, vitamin D, PRP, chitosan formulation to the defect area positively contributed to the repair of bone defect and remarkable clinical improvement. This new delivery approach could be promising for bone healing after surgical operations that cause bone defect to hard tissues.Supporting bone formation in the early period in the repair of maxillofacial bone defects, to shorten the healing period, to complete the loss of function, phonation and aesthetics of the patients as soon as possible and to make the application routine in real life by doing an experiment where the healing of the defect can be seen, to contribute to the acceleration of healing, to help shorten the period of edentulism.
