Aging is the sum of events that lead the organism to death as a result of structural and
functional changes, which are regulated by a genetic program and occur with the effect of
environmental factors. After it was understood that aging is a genetically regulated process,
which genes play a role and how these genes affect aging with environmental changes have
gained great importance. In addition, the detection of metabolic interventions and active
substances that delay or slow down aging paved the way for intervention in the aging process.
Metformin, which has been used as a primary drug in the treatment of type 2 diabetes for
many years, has also started to be investigated as an anti-aging drug because it affects nine
distinguishing features of aging. In this study, we focused on the changes in uncharacterized
genes in the RNA-seq analysis we conducted to investigate which genes resulted from the
change in expression of this life-prolonging effect in fission yeast, whose lifespan was
increased by approximately fifty percent under metformin treatment. In this study, we focused
on the changes in uncharacterized genes in the previous RNA-seq analysis we conducted to
investigate the expression of genes that increased the lifespan of the fission yeast by about
fifty percent under metformin treatment. Among uncharacterized genes that are predicted to
have different functions and whose expression varies at different rates, it was observed that
the expression of genes related to meiosis and genes responsible for recombination increased
as a result of metformin treatment. Among these genes, genes whose expression is strongly
increased may be important candidates for understanding the aging process. In addition, the
upregulation of genes related to meiosis suggests that metformin increased lifespan by
mimicking calorie restriction conditions even in the presence of glucose.
