Özet

ÖZET

Amaç: Yapılan çalışmalarda miR-7-5p’nin tümör supressör olarak birçok kanser türünde rol oynadığı gösterilmiştir. Ancak literatürde miR-7-5p ve hedef genleri olan EGFR ve SKP2’nin AML’deki rolü üzerine herhangi bir çalışma yoktur. Bu amaç doğrultusunda HL60 ve NB4 hücrelerine miR-7-5p’ye ait mimik transfeksiyonu gerçekleştirilmiştir ve proliferasyon üzerindeki etkisi incelenmiştir. Bunun yanı sıra miR-7-5p ve olası hedef genleri SKP2 ve EGFR’nin ifade düzeylerinin AML hücre hatlarında incelenmesi amaçlanmıştır.

Yöntem: Bu çalışmada öncelikle HL60 ve NB4 hücre hatlarına miR-7-5p’ye ait mimik transfeksiyonu yapılmıştır. Transfeksiyonun gerçekleşip gerçekleşmediğinin belirlenmesi için miR-7-5p’nin hücre hatlarındaki ifade düzeyi qRT-PZR yapılarak değerlendirilmiştir. HL60 ve NB4 hücrelerindeki ektopik ekspresyonu artan miR-7-5’nin hücre proliferasyonuna olan etkisi WST 8 yöntemi kullanılarak incelenmiştir. miR-7-5p mimiği transfekte edilen HL60 ve NB4 hücrelerinden önce RNA izole edilerek cDNA’ya dönüşümü sağlanmış ardından qRT-PZR yapılarak SKP2 ve EGFR genlerinin ifade düzeyindeki değişimler qRT-PZR yapılarak belirlenmiştir. Ekspresyon analiz verileri rölatif kantitasyon analizi ile incelenmiştir.

Bulgular: miRNA transfeksiyon validasyonu için yapılan qRT-PZR sonucunda transfeksiyonun başarılı şekilde gerçekleştiği tespit edilmiştir. miR-7-5p transfekte edilen hücrelerde kontrol grubuna kıyasla hücre çoğalmasının anlamlı seviyede azaldığı WST-8 tekniğiyle gösterilmiştir. MiR-7-5p’nin olası hedef geni olan SKP2’nin her iki hücre hattında da mRNA seviyesinde ifadesinin istatistiki olarak anlamlı düzeyde azaldığı belirlenmiştir. EGFR gen ifadesinin ise istatistiki olarak anlamlı şekilde değişmediği tespit edilmiştir.

Sonuç: Çalışmamızda miR-7-5p’nin AML hücre proliferasyonunda rolünün olduğu ve bu miRNA’nın SKP2 genini hedefleyerek genin mRNA ifade seviyesini değiştirebileceği tespit edilmiştir. Tespit ettiğimiz bu bulgunun protein seviyesinde validasyonunun Western-blot yöntemiyle incelenmesi ve miR-7-5p’nin SKP2 genini doğrudan hedefleyip hedeflemediğinin de lusiferaz deneyi ile araştırılması önem taşımaktadır.

ABSTRACT

Aim: miR-7-5p plays critical role as a tumor suppressor in many cancers. However, there is no study in the literature on the role of miR-7-5p and its potential target genes EGFR and SKP2 in AML. For this purpose, miR-7-5p mimic transfection was performed in HL60 and NB4 cells (AML cell lines) and its effect on proliferation was investigated. In addition, it was aimed to examine the expression levels of miR-7-5p and its target genes SKP2 and EGFR.

Methods: miR-7-5p mimic transfection was first performed in HL60 and NB4 cell lines, and the expression level of miR-7-5p in cell lines was evaluated by qRT-PCR in order to confirm whether the transfection occurred. The effect of miR-7-5p on cell proliferation was investigated using the WST-8 method. The expression level of SKP2 and EGFR genes were investigated by qRT-PCR. Expression analysis data calculated with relative quantitation analysis.

Results: As qRT-PCR showed that miRNA transfection was successful. The WST-8 technique demonstradet that cell proliferation was significantly reduced in miR-7-5p transfected cells compared to the control group. It was determined that the expression of SKP2 was statistically downregulated in both cell lines, however EGFR gene expression did not change statistically.

Conclusion: It has been determined that miR-7-5p has a role in AML cell proliferation and this miRNA can change the mRNA expression of SKP2. However, it is important to examine the validation at the protein level by Western-blot method and to investigate whether miR-7-5p directly targets the SKP2 gene with the luciferase assay.
