Fenazopiridin Molekülünün Moleküler Yapı ve Titreşimsel Özellikleri: IR, Raman ve DFT çalışmaları

Fenazopiridin lokal bir anestezik olup, üriner sistem hastalıklarının tedavisinde kullanılmaktadır. Bu molekülün titreşim hareketleri ve ilgili spektral verileri, deneysel IR ve Raman spektroskopik ölçümlerle ve teorik Moleküler Modelleme yöntemleriyle araştırılmıştır. Serbest molekülün belirlenen kararlı iki konformeri için, geometrik parametreler, SCF enerjileri, dipol moment ve termodinamik fonksiyonlar, öncelikle B3LYP/6-31G(d) daha sonra B3LYP/6-311++G(d,p) teori düzeyleri kullanılarak geometri optimizasyonunda hesaplanmıştır. Elde edilen hesaplama sonuçları, her iki konformerde de azo grup, fenil ve piridin halkalarının aynı düzlemde olduğunu açıkça göstermiştir. Bu iki konformerin harmonik ve anharmonic IR ve Raman spektral verileri, geometri optimizasyonunda aynı teori seviyesinde de hesaplanmıştır. Teorik olarak elde edilen temel badlar deneysel olarak gözlenen temel badlarla karşılaştırıldığında, yüksek frekans bölgesindeki anharmonic dalga sayılarının deneysel verilerle tam uyuştuğu, harmonik dalga sayılarının ise ölçeklendirilmesi gerektiği görülmüştür. Bu nedenle harmonik dalga sayıları “İkili Ölçekleme faktörleri”[1,2] olarak adlandırılan yöntem kullanılarak ölçeklenmiştir. Elde edilen teorik spektral verilerin ışığı altında, Fenazopiridin molekülünün deneysel IR ve Raman spektrumlarında gözlenen temel bantlar, başarılı bir şekilde işaretlenmiştir.

Phenazopyridine is a local anesthetic used in urinary tract disorders. The vibrational motions of this molecule and associated spectral data were investigated by means of experimental IR and Raman spectroscopic measurements and theoretical molecular modeling. The resultant equilibrium  geometrical parameters, SCF energies, dipole moment and thermodynamic functions for the determined two stable conformers of the free molecule were calculated by means of geometry optimizations carried out first at B3LYP/6-31G(d) and then at B3LYP/6-311++G(d,p) level of theory.  The obtained calculation results have very clearly indicated that in both conformers, the azo group, phenyl and pyridine rings are coplanar. The harmonic and anharmonic IR and Raman spectral data of these two conformers were also calculated at the same levels of theory as used in the geometry optimizations. A comparison of the obtained theoretical spectral data with those of the fundamental bands observed in the recorded IR and Raman spectra  have showed that the harmonic wavenumbers, in particular those calculated for the high-frequency regions, needs to be scaled properly, though the anharmonic wavenumbers quite well fit to the experimental ones. From this point of view, all the harmonic wavenumbers calculated in this study were empirically scaled by using a direct scaling approach called “dual scale factors” [1,2].  In the light of the obtained theoretical vibrational spectral data, a complete assignment of the fundamental bands observed in the experimental IR and Raman spectra of Phenazopyridine molecule was achieved successfully.
