Amaç Bu çalışmada, Aquilaria agallocha (Thymelaeaceae) odun kısımlarından elde edilen bitkisel yağın 7 farklı konsantrasyonu (% 2, 4, 6, 8, 10, 16 ve 24) hazırlanmıştır. Elde edilen bitkisel yağın akciğer kanseri hücre hattı A549 üzerine etkilerinin araştırılması amaçlanmıştır. Gereç ve Yöntemler Diferansiyel Taramalı Kalorimetre Cihazı (DSC), Yüksek Performanslı Sıvı Kromatografisi (HPLC), xCELLigence, MTT Canlılık testi Aquilaria agallocha’ nın odun kısmından elde edilen numuneler kurutulup toz haline getirildikten sonra yağ ekstraktı elde edildi. Elde edilen yağın içerik analizi HPLC ile yapıldı. Numune ayrıca DSC ile de karakterize edildi. Karakterizasyon aşamaları tamamlanan bitkisel yağın, A549 hücre hattı üzerine etkilerini belirlemek için xCELLigence gerçek zamanlı hücre analiz sisteminde her bir konsantrasyon için olası proliferatif ve sitotoksik etkileri in vitro olarak araştırılmıştır. Sonuçlar, yapılan MTT canlılık testleri ile de uyumlu bulundu. Uygulanan konsantrasyonların etkileri karşılaştırılarak ayrı ayrı değerlendirilmiştir. Bulgular Yağın içeriğinin HPLC sonuçları doğrultusunda; yaklaşık % 32 Omega 6 ve yaklaşık % 20 Omega 9 gibi uzun yağ zincirlerinden oluştuğu saptandı. DSC ile saptanan 95 °C ve 296 °C’deki tepe noktaları, omega 3 ve 6’da tespit edilen β-CD ile uyumlu olarak belirlendi. Gerçek zamanlı xCELLigence hücre analiz sisteminde ise madde uygulandıktan ve MTT canlılık testi yapıldıktan sonra, akciğer kanser hücrelerinin çoğalma ve canlılık değerlerinin zamanla azaldığı görüldü. Sonuç Aquilaria agallocha’dan elde edilen yağın omega 6 ve omega 9 açısından zengin olduğu tespit edildi. Bu maddenin akciğer kanser hücrelerinin çoğalmasını engellediği belirlendi. Yapılacak ileri safha çalışmalar ile ilaç etken maddesi olarak kullanımının uygun olduğu düşünülmektedir.

Effects of Plant Oil from Aquilaria
agallocha on Lung Cancer Cell Line A549 Abstract Aim: In
this study, 7 different concentrations (2, 4, 6, 8, 10, 16 and 24 %) of plant
oil obtained from wood parts of Aquilaria
agallocha (Thymelaeaceae) were prepared. The aim is to investigate the
effects of the oil on the lung cancer cell line A549. Material and Methods: Differential Scanning Calorimetry Device (DSC), HPLC, xCELLigence,
MTT Viability Test The
oil extract was obtained the wood part of Aquilaria agallocha were dried and
powdered. Content analysis of the obtained oil was done by High Performance
Liquid Chromatography (HPLC).
The sample was also characterized by DSC. In order to determine the effects
of vegetable oil on the A549 cell line, the possible proliferative and
cytotoxic effects for each concentration were investigated in the xCELLigence
system. The results were also found to be compatible with the MTT viability
tests. The effects of the concentrations were evaluated separately by
comparing them. Results: According to HPLC results of oil
content in water; it was determined that it consists of long fat chains such
as about 32% Omega 6 and about 20% Omega 9. Peaks at 95 °C and 296 °C
detected by DSC were consistent with β-CD detected in omega 3 and 6. In the
real-time xCELLigence cell analysis system, and the MTT viability test was
performed, it was observed that the proliferation and viability values of
lung cancer cells decreased over time. Conclusion:
It was determined that the oil obtained from Aquilaria agallocha is rich in
omega 6 and omega 9. This substance inhibited the proliferation of lung
cancer cells. It is thought to be suitable for use as an active drug
substance with advanced studies to be done. |
