Özet
Ferulago species, members of the Apiaceae family, are commonly known in Türkiye as “Çakşır” or “Çağşır” and possess significant phytochemical potential due to their rich content of coumarin derivatives. Ferulago species have traditionally been used against hemorrhoids, intestinal worms, ulcers, nausea, and headaches, as well as to facilitate digestion, although they are primarily known for their aphrodisiac properties. In light of these traditional uses, various biological activity studies have demonstrated the cytotoxic, antimicrobial, antioxidant, immunomodulatory, antiangiogenic, and anti-inflammatory properties of Ferulago species [1]. In this study, we investigated the roots of Ferulago antiochia Saya & Miski, an endemic species of Türkiye, to elucidate its phytochemical profile, isolate secondary metabolites, and determine the cytotoxic activities of the isolated compounds. The plant material was continuously extracted with dichloromethane using a Soxhlet apparatus, yielding 3.0 g of crude extract (4.3% yield). The dichloromethane extract was initially fractionated by flash chromatography on a CombiFlash system using a reversed-phase C₁₈ column and a water–acetonitrile gradient as the mobile phase. A total of 66 fractions were collected during the crude fractionation process. Based on their thin-layer chromatography (TLC) profiles, the fractions were combined and further purified using Sephadex LH-20 open-column chromatography and preparative TLC. Structural elucidation was primarily based on 1H-NMR data, supported by SciFinder searches and literature comparisons. Thirteen compounds were isolated and identified as isogosferol, xanthotoxin, psoralen, nodakenetin, oxypeucedanin hydrate, isoimperatorin, pranchimgin, isoscopoletin, umbelliferone, bergaptol, saxalin, falcarindiol, and imperatorin. The cytotoxic activities of the isolated compounds against lung, breast, colorectal, prostate cancer cell lines, and human umbilical cord cell line, will be evaluated using the MTS cell viability assay. Compounds showing prominent cytotoxic activity and/or cancer selectivity will be further evaluated for their effects on apoptosis-related BAX, BCL-2, and caspase-9 protein levels using ELISA. Molecular docking studies will also be performed to investigate their interactions with relevant biological targets.
