KEDİ VE KÖPEK DOKULARININ DÖRT FARKLI ÇÖZELTİ İLE TESPİTİ VE BU ÇÖZELTİLERİN HİSTOPATOLOJİK GÖRÜNÜMLERE ETKİLERİ AÇISINDAN KARŞILASTIRILMASI

Tespit işlemi, histopatolojik olarak incelenmek istenen doku örneklerini stabilize ederek o anki durumlarının görüntülenmesini sağlar. Aynı zamanda; dokunun otolitik enzimler ve mikroorganizmalar aracılığı ile bozulmasını ve morfolojik özelliklerini kaybetmesini önler, şekil ve hacim açısından dokuyu sabitler ve daha sonraki süreçlerde gerekecek başka inceleme yöntemleri için dokuyu muhafaza eder.
Bu çalışmada farklı fiksatif çözeltilerin doku tespitindeki güvenilirliklerini ve aralarındaki farklılıkları ortaya koymak amaçlandı. Bunun için; %4’lük Formaldehit, %10’luk Formaldehit, Hollande ve Carnoy solüsyonları olmak üzere 4 farklı fiksatif tercih edildi. Çalışmada, kedi ve köpek nekropsilerinden alınan 10`ar dalak ve böbrek doku örnekleri her solüsyon için belirtilen bekleme süresi dikkate alınarak bu 4 farklı solüsyonda tespit edildi. Ardından doku örnekleri rutin doku takip işlemlerinden geçirildi ve parafin bloklara gömüldü. Parafin bloklardan 3-4 m kalınlığında kesitler alındı. Kesitler hücrelerin sitoplazmik sınırları, boyanma yetenekleri, nükleer ve nükleolar detayları açısından ışık mikroskobunda incelendi.

THE FIXATION OF THE TISSUES FROM CATS AND DOGS BY FOUR DIFFERENT SOLUTIONS AND THE COMPARISON OF THESE SOLUTIONS BASED ON THEIR HISTOPATHOLOGICAL EFFECTS

The fixation process stabilizes the tissue specimens for histopathological examination in order to show their current state. At the same time; the fixation process inhibits the degradation and morphological deterioration of the tissue by autolytic enzymes and microorganisms, fixes the tissues in terms of shape and volume, and retains the texture for further examination methods that will be required in the subsequent processes.
The aim of this study was to evaluate the reliability and differences between different fixative solutions. With this aim four different fixative solutions; %4 Formaldehyde, %10 Formaldehyde, Hollande’s and Carnoy’s Solutions were preferred. During the study, 10 spleen and kidney tissue specimens from cat and dog necropsies were fixed in these 4 solutions regarding their individual incubation time. After that; the tissue specimens were routinely processed and embedded in the paraffin blocks. Tissue sections approximately 3- 4 µm thickness were cut from the tissue blocks. Sections were examined under light microscope for cytoplasmic margins of cells, staining ability, nuclear and nucleus details.
