Abstract

Triazoles are a variety of heterocyclic compounds which have a five-membered ring of two carbon atoms and three nitrogen atoms. 1,2,4-triazole structure has a spesific importance because of behaving as an important pharmacophore by interacting at the active site of receptor as hydrogen bond acceptor and as a donor. Additionally, because of the polar qualification of the structure, the triazole moiety can increase the solubility of the ligand and by that way increase positively the pharmacokinetic profile of the drug. In literature, structures carrying 1,2,4-triazole-3-thione moiety reported as anticonvulsant, antidepressant, antioxidant, antiinflammatory, analgesic, antinociceptive, antibacterial, antimycobacterial, antifungal, antiviral, anticancer, anti-parasitic, anti-urease agents. Because of the wide pharmacological activity of the compounds carrying 1,2,4-triazole-3-thione moiety, they got attention of the researchers about synthesizing new derivatives of of the compounds carrying 1,2,4-triazole-3-thione moiety which are expected to show better pharmacological activity and to have a better pharmacokinetic profile. We conducted a review study about discovery, synthesis and activity evaluation of new compounds bearing 1,2,4-triazole-3-thione moiety within the last 50 years. And as a result of our literature study, we detected that there are many kind of 1,2,4-triazole-3- thione derivatives that exhibit serious pharmacological activity and possess potential to be a leading compound. Therefore, the researchers definitely should continue their drug discovery investigations and the researches should be supported by molecular modelling studies. Additionnaly as innovation we aimed to provide scientists a wide data resource about 1,2,4- triazole derivatives, thus helping them perform a more organized and fertile drug discovery operation.
