INVESTIGATION OF THE EFFECT OF PENTOXIFYLLINE ON HIF-1ALPHA GENE EXPRESSION AND SERUM HIF1ALPHA LEVELS IN IRRADIATED RATS

Abstract

Introduction The aim of our study is to determine the efcacy of pentoxifylline against hypoxic damage in fbrous tissues, based on the HIF-1α gene expression level, in rats with an irradiation-induced fbrosis model. Methods 17 male Sprague Dawley-type rats used in our study and divided into three groups. The frst group is consisting of rats that received only radiotherapy (RT). In the second group, radiotherapy was administered, followed by oral pentoxifylline for a 4-week period to enhance vascularity (PTX-RT). The third group is the control group (C). Oral pentoxifylline was administered to three rats randomly selected from the control group for 4 weeks (C+). 90 Gy radiotherapy was administered to the legs, targeting the M. Rectus femoris. All rats were sacrifed on the 24th day. Muscle tissues and blood samples were taken. RT- PCR was used to determine the HIF1 alfa gene expression in muscle tissue and HIF-1α ELISA kit was used for the determination of HIF-1α level in serum samples. Statistical analyses were performed with the help of SPSS version 25.0 program. Kolmogorov-Smirnov test, Mann Whitney U Test, and Kruskal Wallis Test was used for analysis. Results There was no signifcant relationship between RT, PTX-RT, C (+) and C (-) groups in terms of ELISA values (p: 0.264). There was no signifcant relationship between RT, PTX-RT, C (+) and C (-) groups in terms of Real-Time PCR values (p: 3,317). Conclusions When the results obtained are considered together with the histopathological analysis, it is concluded that RBF started to develop in a 24-day period with a single fractional 90 Gy dose applied to the rats in our study, but could not develop completely, HIF-1α gene expression values did not show a signifcant variability during the 24-day period and therefore, we cannot determine the efectiveness of pentoxifylline on HIF-1α gene expression through this study
