LPS ile İndüklenen THP-1 Hücre Soyunda Kolşisinin Matriks Metalloproteinaz Aktivitelerine Etkisi

Özet

nflamasyonda sitokin ailesinin üyesi olan interlökinlerin önemli bir rolü bulunmaktadır.
İnflamasyonla ilişkili olduğu bilinen enterobakterial LPS’nin (lipopolisakkarid) IL-1β, TNF-
α, IL-6 gibi öncü-inflamatuar sitokinlerin salınımını TLR (toll benzeri reseptör) ve NF-?β
(nüklear çekirdek faktör) aracılığı ile düzenler. IL-1β’nın, ailevi akdeniz ateşi (FMF), tip 2
diyabet, romatoid arthrit, multipl skleroz ve birçok otoinflamatuar hastalıkta arttığı
bilinmektedir. Matriks metalloproteinazlar (MMP) fizyolojik ve patolojik doku yıkımında
görevli hücre dışı proteazlardır. Kanser gelişiminde rol aldıkları bilinmektedir. Ayrıca matriks
yıkımına neden olan proteazlar olarak birçok kanser tipinde metastazdan sorumludurlar.
MMP’lerinde bazı sitokinlerin aktivasyonunu sağladığı bilinmektedir. Kolşisin, “Güz
Çiğdemi” (Colchicum autumnale) çiçeğinden elde edilen anti inflamatuar bir ilaçtır. Kolşisin
aynı zamanda monosit ve nötrofil kemotaksisini azaltır. Lökositlerde c-AMP düzeyini
arttırarak lizozomal degranülasyonu inhibe eder. FMF tedavisinde atakları önlemede
kullanılmakta ve amiloidoz riskinden de hastayı kurtarmaktadır. Mikrotubül
depolimerleşmesine neden olduğu bilinmekle birlikte etki mekanizması henüz tam olarak
aydınlatılmamıştır. Bu çalışmada 50 ng/ml LPS ile 1 saat süre ile indüklenmiş THP-1 (insan
monositer lösemi) hücrelerinde artan derişimlerde kolşisinin (100, 500, 1000 ng/ml) MMP’ler
üzerine etkisi 12 saat süre sonunda değerlendirildi. Salınan sitokin miktarları SDS-PAGE
sonrası Western Blot yöntemi kullanılarak İmaje J ile analiz edildi. Elde edilen veriler
istatistiksel olarak SPSS Windows 11 ile değerlendirildi. LPS varlığında THP-1 hücrelerinde
MMP aktivitelerinin arttığı izlendi. Farklı derişimlerde kolşisine yanıtın ise doza bağlı olarak
değiştiği ve yüksek dozda sitotoksik etkiye yol açtığı görüldü.

Interleukins that are one of the members of cytokine family play an important role in
inflammation. Enterobacterial LPS (lipopolysaccaride) regulates the release of cytokines with
mediators like TLR (toll like receptor) and NF-?β (nuclear factor kappa-light-chain-enhancer
of activated B cells). It is obvious that the amount of IL-1β induces in FMF (Familial
Mediterranean Fever), type 2 diabetes, romataid arthyrths, multiple sklerosis and most of the
autoinflammation diseases. Matrix metalloproteinases (MMPs) are the outer cell proteases
have a function in the physicolgical and pathological tissue degredation. They play role in the
cancer progress. Moreover, proteases cause matrix degradation in metastasis. MMPs provide
the activation of some cytokines. Colchicine is an anti inflamatuar drug that is extracted from
Colchicum autumnale. It also reduces the monocyte and neutrophile chemotaxis. In
leucocytes, it induces the cAMP level and inhibits the lysosomal degredation. It is used for
preventing the attacks and it protects the patients from amiloidosis. The main effect is known
as microtubule degradation but the mechanism is still unknown. In this present study, MMP
activity was determined in the presence of increasing amounts of colchicine (100, 500, 1000
ng/ml) for 12 hours in THP-1 (Human acute monocytic leukemia cell line) induced with 50
ng/ml LPS for 1 hour. The released cytokine amounts were determined with Western Blot
technique after SDS-PAGE by using İmage J. Data were analysed with SPSS for Windows
11. The induction of MMPs activity in the presence of LPS was observed. The colchicine
response to different concentrations was releated to dose and it was concluded that high doses
of colchicine causes the cytotoxicity.
